Senna leaf extract is an active substance extracted from the leaflets of the legume *Senna angustifolia* or *Senna acuminata*. Its main components are bianthrone glycosides (such as sennosides A, B, and C) and anthraquinone compounds. Its laxative effect stems from the metabolism of sennosides into rhein anthrone by colonic microorganisms, which stimulates intestinal contractions and inhibits water absorption. This extract is used in pharmaceuticals and health supplements for short-term relief of constipation, requiring strict dosage control. It has also been found to regulate colonic protein expression.

Active ingredients
Narrow-leaved senna leaves contain sennoside C, namely rhein-aloe-emodin-diathrone-8,8′-diglucoside. The pods contain sennosides A and B;
Glucosides of rhein and emodin, and trace amounts of emodin or emodin glucoside. Pointed-leaved senna leaves and pods contain 0.85–2.86% and 2.34–3.16% anthracene components, respectively, from which rhein, emodin, small amounts of emodin, and sennosides A, B, and C can be isolated. These anthracene components are all present as glycosides. This plant also contains 3,5-dimethyl-4-methoxybenzoic acid.

The most important markers for standardization are sennosides A and B. They are "prodrugs" that don't do anything until they get to the colon.
Smaller amounts of sennosides C and D are present, but they add to the total impact.
Aloin and rheins are minor derivatives of anthraquinones.
Mucilage and flavonoids are secondary metabolites that have somewhat supporting roles.
Standardization Note: Using HPLC (High-Performance Liquid Chromatography), professional extracts are usually standardized to Sennosides 10%, 20%, or 60%.
Function
- Targeted Activation: Sennosides travel unaltered through the upper gastrointestinal tract. Rheinanthrone, the active form, is released when the sugar molecules are broken down by gut bacteria called bifidobacteria in the colon.
- The myenteric plexus is stimulated to induce muscular contractions (peristalsis), which speeds up fecal transit, by irritating the intestinal mucosa.
- Fluid Accumulation: It softens the stool by increasing the flow of fluids into the intestinal lumen and preventing the large intestine from absorbing water and electrolytes.
- Short-term Relief: Usually results in a bowel movement six to twelve hours after oral ingestion.
